If it works over ethernet via the router then there's a compatibility or setting issue. Try enabling multicasting.

Now I can only get TM to work running 4.1.8 if am on ethernet. Nothing has changed settings wise on the router or ReadyNAS other than the upgrade to 4.1.8. Is multicasting a feature of the router?

Manually connect with finder to the readynas manually (go -> connect to server) and put in the username/pass@ip address (afp://READYNAS:email@example.com) and it mounted the network share. Then from time Machine i was able to select readynas backup and it works. I had no issues with doing this wirelessly or wired. Pre Lion and with 4.1.7 it worked fine wirelessly too so I don't think that is the issue most people are having.
